HEMOCLOT™ Protein S is a clotting-based assay measuring free (functional) Protein S activity in citrated plasma. Principle: APC + patient free PS ? enhanced degradation of FVa ? prolonged clotting time proportional to free PS level. Uses FV-deficient plasma substrate. For thrombophilia screening: PS deficiency (Type I: low total + free PS; Type III: normal total, low free PS) carries 2-3x increased VTE risk. Free PS is the active APC cofactor — total PS includes inactive C4b-BP-bound fraction. Calibrate with Plasma Calibrator (222101). CE-IVD.
