The Xenometrix umuC complete test kit provides all components required for the ISO 13829 umuC SOS induction genotoxicity assay in a 96-well microplate format.
Protocol overview: test strain TA1535/pSK1002 is cultured overnight in LB-ampicillin medium, then exposed to test substance (with and without S9 metabolic activation) for 2 hours at 37 degrees C; SOS induction is quantified as beta-galactosidase activity measured after addition of ONPG substrate, with results expressed as induction factor (IF) relative to the solvent control; genotoxicity is indicated by IF greater than or equal to 1.5 in the absence of cytotoxicity.
Kit components: freeze-dried TA1535/pSK1002 tester strain with revival protocol; LB broth with ampicillin (200 mg/mL ampicillin stock); ONPG (o-nitrophenyl-beta-D-galactopyranoside) substrate solution; 4-nitroquinoline-1-oxide (4-NQO) positive control (direct-acting); 2-aminoanthracene (2-AA) positive control (S9-dependent); sodium phosphate lysis buffer; Z buffer with beta-mercaptoethanol.
