XenoScreen YES (Yeast Estrogen Screen) and XenoScreen YAS (Yeast Androgen Screen) from Xenometrix are yeast-based cell reporter assays for quantitative detection of estrogenic and androgenic (or anti-estrogenic/anti-androgenic) activity in environmental water samples, cosmetic ingredients, industrial chemicals, and pharmaceutical compounds. Both assays use recombinant Saccharomyces cerevisiae strains expressing the human estrogen receptor alpha (YES) or human androgen receptor (YAS) fused to a beta-galactosidase reporter gene.
Binding of estrogenic or androgenic compounds to the receptor activates transcription of the lacZ reporter, producing measurable beta-galactosidase activity quantified colorimetrically with ONPG substrate.
XenoScreen YES and YAS are validated per ISO 19040-1 (Water quality --- Determination of the estrogenic potential of water and waste water) and are accepted by regulatory agencies for endocrine disruption screening under REACH, EU Water Framework Directive, and OECD Conceptual Framework for endocrine disruptor testing.
