2-Aminoanthracene is a polycyclic aromatic amine activated to its mutagenic hydroxylamine form by cytochrome P450 enzymes in rat liver S9 fraction. A positive mutagenic response to 2-AA confirms that the S9 metabolic activation system is functional. It is the standard positive control for validating S9 quality in Ames and genotoxicity assays per OECD 471 requirements.
