TA1535[pSK1002] combines base-pair substitution detection (TA1535 histidine mutation) with SOS genotoxicity reporting via the umuC::lacZ fusion on the pSK1002 plasmid. Upon exposure to genotoxic chemicals, the umuC gene is induced as part of the SOS DNA damage response. The pSK1002 plasmid carries a umuC–lacZ gene fusion; SOS induction is quantified by measuring β-galactosidase activity via conversion of ONPG (o-nitrophenyl-β-D-galactopyranoside) to yellow o-nitrophenol at 420 nm.
