2-Aminofluorene requires S9 metabolic activation to the mutagenic N-hydroxy form. It induces both frameshift mutations (detected by TA98) and base-pair substitutions (detected by TA100), making it a dual-activity S9-dependent positive control that simultaneously validates S9 function across the two most important OECD 471 tester strains.
