The OECD TG 471 metabolic activation system uses a combination of rat liver S9 fraction and a co-factor solution to reconstitute cytochrome P450 enzyme activity ex vivo — providing the Phase I and Phase II metabolic enzyme activity required to convert indirect-acting promutagens and progenotoxins to their reactive metabolites.
The S9 mix is prepared by combining S9 fraction with co-factor solution immediately before use; the co-factor solution provides MgCl2 (cofactor for CYP enzyme activity), KCl (ionic strength control), glucose-6-phosphate (NADPH regeneration substrate for the pentose phosphate pathway), NADP+ (cytochrome P450 electron acceptor), and sodium phosphate buffer pH 7.4.
Xenometrix S9 co-factor solution is supplied as a validated, ready-to-use formulation at the exact concentrations specified in OECD TG 471 for standard S9 mix preparation — typically S9 fraction at 10% final volume (v/v) in complete S9 mix.
